Origin: This strain was made by electroporation of DAc8 embryonic stem cells with a targeting vector containing 6.7kb 5 prime and 1.6- kb 3 prime homology arms and a CAG-EGFP-IRES-Pac cassette. Chimeras were formed by microinjecting F344 blastocysts. Founder animals were mated with SD rats.

strain_characteristics Absence of Tp53 protein was confirmed by western blot in homozygous animals. Homozygotes die at an early age; most will develop neoplasia by 4 months of age. Heterozygous animals may also develop neoplasia. Very little clinical signs are seen prior to death and twice a day monitoring maybe needed to detect animals with neoplastic disease symptoms prior to death. Most clinical findings are related to acute blood loss from hemorrhage (hemangiosarcomas are common) or seizure activity/neurologic signs due to brain or spinal cord tumors. 2292495
